Spectrin is an actin binding protein that is a major component of the plasma membrane skeleton. Spectrins function as membrane organizers and stabilizers by forming dimers, tetramers and higher polymers. Vertebrate spectrins have two alpha-subunits (alpha-I/alpha-II), four beta-subunits (beta-I-beta-IV) and a beta-H subunit creating diversity and specialization of function. Spectrin alpha and spectrin beta are present in erythrocytes, whereas spectrin alpha II (also designated fodrin alpha) and spectrin beta I (also designated fodrin beta) are present in other somatic cells. The spectrin tetramers in erythrocytes act as barriers to lateral diffusion, but spectrin dimers seem to lack this function. Spectrin beta III is highly homologous to both spectrin beta I and spectrin beta II. VISTA / GI24 is a transmembrane protein expressed in bone, on embryonic stem cells (ESCs), and on tumor cell surfaces. On ESC s, Gi24 appears to positively interact with BMP-4, potentiating BMP signaling and the transition from an undifferentiated to a differentiated state. On tumor cells, Gi24 both promotes MT1-MMP expression and activity and serves as a substrate for MT1-MMP. This increases the potential for cell motility. Mature human Gi24 contains a 162aa extracellular region with one V-type Ig-like domain and a 96aa cytoplasmic domain. Human Gi24 undergoes proteolytic cleavage by MT1-MMP, generating a soluble 30 kDa extracellular fragment plus a 25-30 kDa membrane-bound fragment. The protein encoded by this gene is a homeobox-containing developmental regulator associated with liver development. The encoded protein binds to the alpha-fetoprotein (AFP) gene promoter and increases the expression of AFP. This gene is overexpressed in some lung cancers and is linked to poor patient survival, possibly due to its resistance to cisplatin. This gene is aberrantly methylated in pancreatic cancer, deleted in squamous cell lung carcinomas, and acts as a tumor suppressor in esophageal cancer. This MAb is specific to lambda light chain of immunoglobulin and shows no cross-reaction with lambda light chain or any of the five heavy chains. Immunoglobulins are four-chain, Y-shaped, monomeric structures comprised of two identical heavy chains and two identical light chains held together through interchain disulfide bonds. The chains form two domains, the Fab (antigen binding) fragment and the Fc (constant) fragment. Immunoglobulin D (IgD) exists as a monomer withδ heavy chains and either κ or λ light chains. It plays a biological role as a transmembrane receptor molecule, co-expressed with IgM on the surface of mature/naive B cells. In particular, it is found on spleen B cell surfaces. Compared to IgM, IgD exists in much lower numbers and is not expressed on immature B cells. IgD surface expression on B cells is regulated in part by IL-27.
